Th1L07 Competitive and
non-competitive inhibitors
- Competitive inhibitors
- block the enzyme active site
- Alter the apparent Km not
the Vmax
- Increase the [S] it takes to
reach Vmax
- Reduces the steepness of the
slope
- E.g. Malonate
- Inhibits succinate dehydrogenase
- Takes up the active site, blocking succinate (the substrate) from entering
the succinate dehydrogenase
- Non-competitive
- reversible or irreversible
- cannot reach the same Vmax
- Km and Vmax/2 is the same
- Does not directly block the active site
- Interfere in some other way with the
catalytic mechanisms
- irreversibly poisons the enzyme
- e.g. chealator (EDTA)
- Reversible non-competitive
enzyme inhibitor
- Inhibits Mg2+ requiring enzyme
