Describe a method of measuring the levels of ATPase in different muscle fiber types. ATPase is pH sensitive. We put the fiber into either alkaline or acid conditions. Fast myosin is active in alkaline solution so the ATPase is active and therefore will take up the fluorescent dye we place in the solution. The darker the dye the greater the activity of ATPase and therefore we can determine fiber type.
