  1. Polymerase Chain Reaction
    1. amplifies the amount of a DNA sample
      1. 1) DNA heated to 95 to break H2 bonds
        1. 2) Cool to 35 to primers bind to start of each strand, prevent rejoining + act as marker for polymerase
          1. 3) heated to 73 + thermostable TAQ polymerase replicates strand
            1. each time process repeats, DNA doubles
      2. DNA probe
        1. locates section of DNA, consists of short lengths of single stranded DNA with specific nucleotide sequence. To dectect- radioactive/fluorescent for xray/laser scanner
          1. DNA profiling ( fingerprinting)
            1. 1) DNA cut with restriction endonucleases into fragments
              1. 2) Fragments seperated by gel electrophoresis due to bands, smaller fragments move further
                1. 3) Bonds heated to be single stranded, transferred onto nylon and washed with DNA probes complementary to MRS's
                  1. 4) Repeated with several probes for different MRS's + fragments detected
                    1. 5) DNA bound to probe show up, bands are fingerprints
