What is chromatography used for? To separate components in a mixture Once separated, each component can be identified
What is the basic set up of chromatography Mobile phase (where molecules can move) Stationary phase (where molecules cannot move)
What is the mobile phase in paper and TLC? Liquid solvent (e.g. ethanol or water)
What is the stationary phase in paper chromatography? A piece of chromatography paper
What is the stationary phase in TLC? Thin layer of solid (e.g. silica gel, on a glass or plastic plate)
What is the basic principle of chromatography? 1. Mobile phase moves through or over stationary phase 2. Components spend different amounts of time in mobile phase and stationary phase 3. Components that spend longer in the mobile phase travel faster/further 4. Time spent in different phases is what separates out the components
How is the Rf value calculated? Rf value = Distance travelled by spot / Distance travelled by solvent
