Questão Responda
Identify the enzyme involved in PCR DNA polymerase is adapted to function at high temperatures without denaturation. It stems from thermophilic microorganisms.
Identify and describe the 1st stage of PCR DENATURATION Extremely high temperatures (approx 95degreesC) breaks the hydrogen bonds between the base pairs on the antiparallel strands.
Identify and describe the 2nd stage of PCR ANNEALING Rapid cooling occurs (approx 55degreesC) allowing specific DNA primers to attach and fix (anneal) to the complementary sequence within the DNA template.
Identify and describe the 3rd stage of PCR ELONGATION The polymerase mixture is heated again to 72 degrees C. The enzyme DNA polymerase (which is not itself denatured by the high temperatures) extends the primers by adding the nucleotides in 5' - 3' direction.
