What are the conditions needed for cell fractionation, and why? Ice cold to reduce enzyme activity Buffered to keep a constant pH In isotonic solution to prevent damage to the cell through osmosis
What is the first step of cell fractionation? Homogenisation- break up the cells
What is the second step of cell fractionation? Filtration- Filter the homogenised cell solution to remove large cell debris or tissue debris
What is the third step in cell fractionation? Ultracentrifugation- Put the solution into a centrifuge. Once done, drain off the supernatant into another tube and spin it again. You can discard the pellet
In what order do organelles separate at? Nuclei, chloroplasts, mitochondria, lysosomes, endoplasmic reticulum, ribosomes
