  1. Smooth Muscle
    1. Spontaneous contractile activity
      1. Contracts Slowly
        1. Not Prone to muscle fatigue
          1. Ideal for sphincter muscles
          2. Destrusor
            1. Contraction mediated by muscarinic receptors
              1. Stimulation by acetlycholine
                1. Inhibited by antagonist atropine
            2. Tissue
              1. Maintained in an aerated physiological solution
                1. Respond to chemical,electrical,mechanical stimui
                  1. continue for many hours for application of many drugs.
                2. Clear quantitive relationship between tissue response and applied drug contraction
                  1. Mesurment recorded by force displacement transducter.
                  2. Set up
                    1. Equiptment
                      1. Power lab 26T
                        1. Micropositioner
                          1. Force Transducer
                            1. Calibrated to give more meaningful units of grams
                              1. Residual offset voltage needs to be corrected
                              2. Ring Stand
                            2. Tissue Preparation
                              1. 2cm strip of bladder
                                1. Keep tissue moist with phosphate buffer
                                  1. Additional 95% O2
                                    1. 5% CO2
                                    2. Attach string to each end
                                      1. Attach to mounting hook, then ring stand
                                        1. Force Tranducer at 1-1.5g tension
                                          1. Submerge in sailine
                                            1. Allow 30minutes to equilibrate
                                        2. Acetylcholine
                                          1. Stock Solution of 10-2 M
                                            1. Serially dilution in 10ml saline (Tyrode Ringer)
                                              1. To give: 10-3.10-4 and 10-5
                                            2. Drug Addition Cycle
                                              1. Wait 3 Minutes to record any response
                                                1. Empty bath and wash the tissue 2 times after recording
                                                  1. Wait additional 3 mins before adding next concentration of drug
                                              2. Acetylcholine Anatgonist
